Equipment details This system is based on a fully motorised Leica DMi8CEL (inverted) research microscope, and the scanhead is equipped with one conventional photo multiplier tube and two highly sensitive HyD detectors. The entire microscope is controlled via the Leica LAS X software , which includes the Navigator module that is very intuitive and ideal for semi-automated image acquisition from large sample areas. The software also provides modules for spectral unmixing and ratiometric Förster Resonance Energy Transfer (FRET). The system is also equipped with a stage top environment control chamber to maintain optimal conditions for live cell imaging (temperature, carbon dioxide concentration and air humidity). Lasers The TCS SP8 provides the following lasers with the specified laser lines for fluorophore excitation: Laser lines [nm] Blue diode 405 Arg 458 488 514 Diode 561 HeNe 633 Objectives The Leica TCS SP8 is equipped with the following lenses: Lens magnification immersion NA WD HC PL APO 10x CS2 10x A 0.4 2.2 HC PL APO 20x CS2 20x A 0.75 0.62 HC PL APO 40x CS2 40x O 1.3 0.24 HC PL APO 63x CS2 63x O 1.4 0.14 HC PL APO 63x 63x W 1.2 W = water, A = air, O = oil, WD = working distance in mm Filter sets for epifluorescence illumination/visual check Standard filter cubes for epifluorescence detection of DAPI (exc 360/40, em LP425), FITC (exc 470/40, em LP515) and Rhodamine (exc 540/45, em LP590) are available on the TCS SP8.
